ABSTRAK

Hipertermofil adalah kelompok mikroorganisme yang mampu hidup pada suhu di atas 70oC bahkan dapat hidup sampai suhu 113oC. Mikroorganisme termofilik dan hipertermofilik memiliki potensi besar sebagai penghasil biokatalis tahan panas. Penelitian ini bertujuan untuk melakukan karakterisasi urutan basa 16S rRNA dari isolat TS3 yang berasal dari Kawah Domas Tangkuban Perahu, Jawa Barat. Isolat ditumbuhkan dalam medium cair Luria – Bertani (LB) yang dimodifikasi pada suhu inbubasi 70oC selama 3 x 24 jam. Penentuan urutan basa 16S rDNA dilakukan dengan hasil amplifikasi gen 16S rDNA diberi label menggunakan Big Dye Terminator Reaction Mixture Sequensing Kit. Isolat diidentifikasi dengan analisis urutan nukleotida 16 S rDNA. Berdasarkan analisis urutan nukleotida dapat disimpulkan bahwa isolate TS3 memiliki kemiripan 80% dengan urutan nukleotida 16S rDNA dengan Arkaebakteria uncultured.

ABSTRACT

Hyperthermophyl microorganism are microorganisms which are able to live at a temperature higher than 70oC and on even up to 113oC. Thermophylic and hyperthermophylic microorganisms have a potency to be used as a heat resistant biocatalyst producers. The aim of this study was to sequencing 16S rDNA from a hyperthermophylic microorganisms isolated TS3 from Domas Crater, Tangkuban Perahu., West of Java. The isolated microorganisms was cultivated in liquid modified Luria-Bertani medium (LB) at 70oC and incubated for 3 x 24 hours. The isolated microorganisms was identified by the sequencing its 16S rDNA. Base on the nucleotide sequence, the isolate wan concluded to be an Archaeabacteria which has a 80% similarity in its 16S rDNA sequence to member of the uncultured Archaeabacteri.
